چکیده

The fact that colchicine inhibits hepatic secretion of very low density lipoprotein (VLDL) particles has been interpreted to mean that microtubules are involved in hepatic V L D L secretion. To further define this relationship, we have at tempted to see if changes in hepatic V L D L secretion are associated with changes in hepatocyte microtubule or tubulin content. Accordingly, hepatic secretion of V L D L was increased in rats, and the hepatocyte content of both microtubules (using quantitative morphometric methods) and tubulin (using a time-decay colchicine binding assay) was determined. In acute experiments, V L D L secretion was increased by perfusion of isolated rat livers for 2 h with varying concentrations of free fatty acids (FFA). Results indicate that hepatic V L D L triglyceride (TG) secretion at perfusate FFA levels of 0.7 /zEq/ml is threefold greater (P < 0.01) than when livers are perfused without added FFA. However, no differences are observed in the content of microtubules in these livers: specifically, microtubules occupy 0.029% of hepatocyte cytoplasm in livers perfused without FFA and 0.030% of cytoplasm in livers perfused with FFA. In chronic experiments, rats were fed for 1 wk with either standard rat chow or a hyperlipidemic (sucrose/ lard) diet. With the experimental diet, plasma triglyceride levels increase threefold over controls, and liver V L D L T G production, as determined by [3H]glycerol turnover studies, is 55% greater (P < 0.01) than controls. However, microtubules occupy 0.027% of the cytoplasm of hepatocyte cytoplasm whether rats are on standard or hyperlipidemic diets. Furthermore, the tubulin content of isolated hepatocytes does change, and represents 1% of hepatocyte soluble protein, irrespective of diet. These results suggest that increases in hepatic V L D L secretion can occur without any demonstrable change in hepatocyte assembled microtubule or tubulin content, and raise questions as to the role played by microtubules in hepatic V L D L secretion.

Evidence that microtubules play a permissive role in hepatocyte very low density lipoprotein secretion

To determine whether a minimum number of assembled microtubules is required for very low density lipoprotein (VLDL) triglyceride TG) secretion in hepatocytes, antimicrotubule drugs of different concentrations were given to rats. Hepatic VLDL-TG release was subsequently measured by a liver perfusion system, and hepatocyte ultrastructural changes were analyzed by quantitative ultrastructural meth...

Inhibition of acyl-CoA: cholesterol acyltransferase decreases apolipoprotein B-100-containing lipoprotein secretion from HepG2 cells.

There is evidence that the overproduction of apoB-100-containing lipoproteins by the liver is the underlying event in some forms of dyslipoproteinemia. This metabolic status is associated to an increased risk of developing premature coronary artery disease CAD. The conclusions from previous studies suggested that the availability to the hepatocytes of cholesterol that is readily esterified is a...

مهار ترشح تری اسیل گلیسرول وابسته به فراکسیون VLDL توسط اگونیست های آلفا- و بتا- ادرنوسپور در هپاتوسیت تهیه شده از کبد رت (Rat)

Background and purpose: Ât hunge state liver is the main source of synthesizing and secretion of low Density Lipoprotein (KLDL). For this purpose triacyleglycerol, phospholipids, cholestrol and its esters must be added to newly synthesized Âpo-B and secreted as a newly synthesized VLDL.VLDH metabolism is under hormonal and metabolic control, and stimulation of both ways is dependent on calciu...

Expression of Endoderm and Hepatic Specific Genes after in vitro Differentiation of Human Embryonic Stem Cells

Background: Human embryonic stem cells (hESC), which are derived from the inner cell mass of the blastocysts, have been considered to be pluripotent cells. In this study we examine the differentiating potential of hESC into hepatocytes by characterization of the expression of endoderm and liver-specific genes. Methods: hESC were cultivated in suspension to form aggregates, the embryoid bodies. ...
